Über den Vortragenden: Julien Béthune leitet die Arbeitsgruppe Molekularbiologie und Zellkulturtechnik an der Hochschule für Angewandte Wissenschaften Hamburg.

Inhalt des Vortrags: Since its initial description in 2012, the proximity-dependent biotinylation technique BioID, coupled to mass spectrometry analysis, has established itself as a robust and viable alternative to the classical pulldown approaches for the identification of protein-protein interactions. Recently we engineering split-BioID, a protein-fragments complementation assay that greatly enhances the resolution of BioID by allowing the analysis of context-dependent protein complexes. We are further developing more versatile proximity-dependent proteomics assays with the goal to obtain smaller labeling enzymes with improved labeling kinetics and that can be used in a split assay format. In this presentation I will showcase ultraID, our latest enzyme, and show how it helped us understand the membrane-associated interactome of the COPI coat protein complex in living cells.
