Metabolic reprogramming is a fundamental mechanism through which cells regulate their fate decisions and adapt to environmental changes. Immune cells, in particular, exemplify this necessity by rapidly adjusting their metabolic profiles to fulfill diverse functional roles. Although it is increasingly evident that metabolic switches are orchestrated through interactions among cellular organelles, our current understanding remains limited to individual organelles or isolated two-way interactions. A comprehensive, holistic insight into how the entire organelle network—termed the organellome—coordinates to regulate cellular metabolism and immune cell fate is notably lacking. This knowledge gap is particularly critical, as dysfunctions within the organelle communication network may underpin various genetic and acquired immune disorders. Our laboratory applies integrative systems biology approaches to systematically define spatiotemporal organelle interaction strategies, aiming to uncover fundamental communication principles of the organellome. By building a functional organelle map, we aim to identify how coordinated organelle networks instruct metabolic decisions to pave the way for novel therapeutic strategies targeting immune-related diseases.
