Inhalt des Vortrags: A couple of years ago, we co-discovered a new gene-regulatory mechanism: Endogenous translational readthrough (stop codon suppression) leads to the expression of ‘extended’ forms of lactate dehydrogenase and malate dehydrogenase. The extensions carry targeting signals for the peroxisome, so that a fraction of these enzymes is imported into peroxisomes, giving rise to a – hypothetical – metabolite shuttle at the peroxisomal membrane [1–3]. In the first part of the talk I will present this discovery; in the second part I will turn to our recent and unpublished work on how we use mouse models and human induced pluripotent stem cells to study the role of peroxisomes in the heart.
